Pkd1 expression in the mouse embryo

Figure 4. Pkd1 expression in the mouse embryo. A heterozygous Pkd1del17-22bgeo E12.5 mouse embryo was stained with X-gal to demonstrate the expression of Pkd1 using a ‘knocked-in’ b-galactosidase reporter gene and imaged using optical projection tomography (Ref. 31). The b-galactosidase reporter gene was fused to the Pkd1 gene so that Pkd1 expression resulted in b-galactosidase expression. The b-galactosidase substrate, X-gal, which produces a coloured product, can then be used to monitor Pkd1 expression. Pkd1 expression (green) is seen in the developing cartilage, cardiac outflow tract, smooth muscle and urogenital sinus in addition to many other tissues. Sections are taken at intervals through a whole-mount embryo to demonstrate the different sites of expression. See also the optical projection tomography of this figure (Movie 1).
