Figure 1. Pathways from glucose entry to extracellular matrix production in cells with increased GLUT1 expression. The figure shows known (solid lines) and proposed (dashed lines) pathways from glucose entry to extracellular matrix (ECM) production in mesangial cells with increased GLUT1 expression. Mesangial cells with overexpression of GLUT1 have been shown to have increased flux of glucose through the aldose reductase and glycolytic pathways. As a result, they demonstrate persistently activated PKCa and PKCb1, resulting in increased AP-1. The AP-1 transcriptional activator then leads to increased ECM production. Sorbitol and lactic acid are produced in excess.
