Figure 1. Methylation and subsequent inactivation of one of the parental alleles of an imprinted gene. In this example, the maternal allele is imprinted. The methylated, or silenced, maternal allele is shown in red, and the active, unmethylated allele in green. The differentially methylated domain (DMD) (grey rectangle) is the element regulating the specific parent-of-origin transcription/expression. In this example, the DMD is located in the promoter region of the gene. DNA methylation inhibits the binding of the transcription complex (TC).
